A functional genomics approach will be used to investigate the cellular mechanisms underlying adverse drug reactions. The focus will be on the kidney, an organ where adverse drug reactions are frequently observed and remain a serious problem in the clinic and for the pharmaceutical industry. An advanced in vitro cell-based model will be used, whereby renal cells are cultured in 3D and undergo morphogenesis, producing multicellular kidney organoids. These structures have a high level of differentiation and organization approaching that seen in the kidney, and therefore serve as a more relevant in vitro model for studying kidney toxicity. The responses of kidney organoids to reno-toxic pharmaceuticals will be determined by gene profiling and automated image analysis of the kidney organoids. Using RNAi-mediated knockdown of genes which are up- and down-regulated in response to pharmaceutical treatment will establish whether those specific genes play a functional role in the stress response. Using this approach, we expect to identify and then further characterize proteins and signalling pathways which mediate the adverse response to pharmaceuticals.
Requirements
Required education/skills:University Graduate
We seek a highly motivated and talented biologist with an interest in cell behaviour and signalling. Candidates should have good communication skills and have a background in cell biology, molecular biology and/or biochemistry and/or bioinformatics.
