Job description
Function title: PhD position Molecular marker discovery and application in onion and leek
The project is part of the Technological Top Institute Green Genetics. The major aim of this challenging research project is to identify Single Nucleotide Polymorphims (SNP) and microsatellite markers associated with resistance in onion (Allium cepa L.) to fungal diseases. In addition, comparative genomic studies will be performed between onion and leek (Allium porrum L.). The project will result in the development of marker based breeding programs aimed at the development of healthier crops, higher yields and sustainable production.
The PhD student will start with the production of a large dataset of approximately 80,000 Expressed Sequence Tag (EST) sequences from cDNA libraries originating from a number of selected genotypes from onion and leek. High through put sequencing facilities will be used to reach this goal. EST sequences and microsatellite sequences will be annotated based on sequence similarity, and collected in a searchable database. SNPs will be discovered in datasets using in-house developed software.
The second aim of the project is to identify associations with genes segregating for resistance to Botryits squamosa (onion-leaf blight) and Fuarium oxysporum (fusarium basal rot). Wild Allium species with resistance to these fungal diseases have been identified and were successfully crossed with onion. The PhD candidate will use available mapping populations. Disease screenings will be carried out by the breeding companies in close collaboration with the PhD candidate and scientists of Wageningen UR Plant Breeding. A collection of SNPs and SSRs will be converted to applicable marker assays and placed on the genetic map of the population.

Required education/skills:University Graduate
We are looking for an enthusiastic and result-driven person with:
• a university degree at MSc level in a relevant field (e.g. plant breeding, genetics or molecular biology)
• experience with (several of) the following techniques: (molecular) genetics, bioinformatics, molecular biology. Affinity with plant-fungal interactions will be of additional value
• an interest in multidisciplinary approaches
• proficiency in and affinity with working with computers and laboratory equipment
• team player with good communication skills.

Estimated maximum salary per month: € 2000 in the first year rising to € 2558 in the fourth year. Duration of the contract: for a period of 18 months with a possible extension of 30 months.
